    Methotrexate or Plaquenil? DailyStrength I do know that plaquenil is generally considered the least invasive drug, generally prescribed as the first line of treatment. Methotrexate is the most commonly prescribed DMARD, but it does come with the chance of side-effects. I never had any side-effects of dire consequence, but many on this forum have.
